• Novel 3D-printedMOF-hydrogel microarray (3D-MHMA) for multiplexed miRNA detection. • The platform integrates MOF-based enrichment/quenching with a CHA-HCR cascade for ultrasensitive, enzyme-free amplification. • We first validated miR-194 as an early-warning biomarker with miR-122 for herb-induced liver injury (HILI). • Platform demonstrated superior early diagnostic capability in vivo and in vitro.
