Here we introduce a micropore array-based single-cell operating system (MASCOS), which realized flexible and efficient single-cell capture, transferring, dropletization, imaging, reaction and analysis. Using a micropore array membrane, the MASCOS can rapidly capture single cells on the micropores under low hydraulic pressure (similar to 12 Pa). A single-cell array was formed on the membrane, ensuring simplified and automated single-cell addressing. On this basis, complex and multistep single-cell operation with high precision could be achieved by programmatically performing the capillary probe-based aspirating-depositing and the moving of an x-y-z translation stage. On the MASCOS platform, every cell in the single-cell array can be individually transferred, then encapsulated by a nanoliter-scale droplet with a success rate of up to 100%. Finally, the feasibility of the present system for single-cell analysis was demonstrated in a single-cell viability assay.
