• A novel class of RNA fluorescent aptamer ligand was developed based on the Pepper aptamer system. • Investigation of the binding behavior of the modified HBC530 with the Pepper aptamer. • CHA-Me/Pepper system can be used to measure and image endogenous hCES1 in living cells. • Induction of inflammation in FLS cells revealed the presence of lipid deposition along with an increase in hCES1. • Revealed abnormal elevation of hCES1 in the RA patient's knee joint synovial fluid for the first time.
