• Patients with non-inflammatory pruritus (NIP) showed zinc deficiency (ZD), which was more pronounced than those with inflammatory pruritus (IP). • Zinc supplementation improved NIP symptom along with normalization of zinc levels. • Compared to control mice, ZD model mice showed an increased sensitivity to morphine-mediated pruritus, which were reversed by kappa opioid receptor agonist, nalfurafine, pretreatment. • The results implicated that zinc is critically related to pruritus through modulation of opioid receptor signaling.
