Acetaminophen is an effective mild analgesic and antipyretic agent. In double-blind, controlled experimental pain studies [animal and human] of short duration it was superior to placebo and produced analgesia comparable to that produced by aspirin. The frequency of adverse reactions to therapeutic doses is low as that of aspirin. Acetaminophen overdosage may result in irreversible hepatotoxicity. Since clinical intoxication manifestations can be of slow onset there may be a tendency to delay initiation of definitive therapy. I.v. cysteamine and possibly oral methionine appear to be effective in preventing hepatotoxicity if administered within 10 h of drug ingestion. Physicians should be aware of the potential danger of overdosage and alerted to its clinical manifestations.
