• Background information on the PI3K-AKT pathway and its relevance to adiponectin and macrophage inflammation has been added to improve logical flow. • Detailed experimental methods (e.g., modeling approach, reagent concentrations, drug doses, gating strategy for macrophages, and transcriptomic analysis) have been provided to enhance reproducibility. • HE staining and immunofluorescence results of adipose tissue from obese mice have been supplemented to highlight mechanistic differences in insulin resistance between CIH and obese mice. • The top 100 differentially expressed genes from transcriptomic sequencing are presented, with PI3K-AKT pathway–related genes identified and validated by qPCR. • The Discussion has been expanded with literature linking adiponectin, macrophage inflammation, PI3K-AKT signaling, and insulin resistance, while noting the lack of knockout/inhibition experiments as a study limitation. • The entire manuscript has undergone language refinement, and page numbers have been added to facilitate review.
