• Stable 5.7 at% Ag HEN coating showed the best overall performance. • The coating strongly inhibited F. nucleatum and P. gingivalis. • S2 enhanced osteoblast differentiation and matrix mineralization in vitro. • S2 promoted bone regeneration and osseointegration in rabbit tibiae. • MAPK/ERK pathway activation mediated the pro-osteogenic effect of S2.
