Abstract

• A self-assembled azoreductase-responsive nano-prodrug was developed for oral IBD treatment. • A-MPD showed high gastrointestinal stability and resisted premature intestinal degradation. • Negative surface charge promoted selective retention at positively charged inflamed colon sites. • Azoreductase-triggered cleavage enabled on-site mesalamine release in the diseased colon. • A-MPD delivered both potent therapy and prophylactic protection against DSS-induced colitis.
