AQbD Driven RP-HPLC method for estimating rizatriptan benzoate and meloxicam: integrating green and blue analytical approaches

The present study aimed to develop and validate a robust, stability-indicating RP-HPLC method for the simultaneous determination of Rizatriptan Benzoate and Meloxicam using an Analytical Quality by Design approach integrated with green analytical principles. A central composite design is used to evaluate critical method variables, such as flow rate and the percentage of organic phase. Separation was achieved on a Shimpack C18 column (250 × 4.6 mm, 5 μm) using methanol and 0.1% formic acid (80:20, v/v) as the mobile phase at a flow rate of 1.0 mL/min, with a detection wavelength of 296 nm. The method was validated in accordance with ICH guidelines for system suitability, linearity, precision, accuracy, robustness, specificity, sensitivity, and forced degradation studies. Greenness was assessed using AGREE, MoGAPI, and an Analytical Eco-Scale. Blue applicability grade index implemented to evaluate the practicality of a method in routine analysis. The optimized method showed retention times of 1.89 min for Rizatriptan Benzoate and 7.97 min for Meloxicam with a resolution of 23.3. Linearity was observed over the 5–50 µg/mL range, with correlation coefficients of 0.999 for Rizatriptan Benzoate and Meloxicam. The percentage recovery values ranged from 100.35% to 100.71%, and the percentage RSD for precision ranged from 0.62% to 0.98%, confirming the accuracy and precision of the method. The method also showed good robustness and specificity. The LOD values for RZT and MLX were 0.07 µg/mL and 0.19 µg/mL, respectively, while the LOQ values were 0.20 µg/mL and 0. 50 µg/mL, respectively. Green evaluation scores from AGREE (0.70), MoGAPI (80), and Analytical Eco Scale (77) confirmed the method as eco-friendly. The BAGI score of 80 ensures the practicality for routine use. The proposed method is simple, reliable, robust, and sustainable for the simultaneous analysis of Rizatriptan Benzoate and Meloxicam.
