Redox proteomics has revealed thousands of oxidative post-translational modifications across the proteome. The functional consequences of most oxidative post-translational modifications remain unknown. Fold change-based ranking enriches for highly reactive cysteines but can miss low-stoichiometry allosteric redox switches. Functional genomic screens provide an orthogonal approach to deconvolute the importance of redox-active sites identified in proteomic analysis.
