• SS-OCTA reveals spatial heterogeneity in choroidal thickness and perfusion in guinea pigs. • The C1–6 mm and superior regions exhibit heightened sensitivity to FD stimuli. • Brimonidine inhibits myopia by preserving choroidal thickness and perfusion. • Brimonidine prevents myopia via a choroidal mechanism involving NOS and TGF-β1.
