• Engineering of Protein G for affinity capture of antibodies via interaction with the Fab-fragment. • Affinity purification of full-length monoclonal mouse antibodies is presented. • Mild elution from the resin at neutral pH (7.0) is possible. • The structure-function relationship of the calcium-dependent elution mechanism is described. • A valuable tool for purification of acid-sensitive antibodies and Fab-fragments is developed.
