• Chronic exposure to chlorate induces neuronal senescence at regulatory safety limits. • Sub-cytotoxic exposure activates p21/p16-mediated aging in human iPSC-derived DAn. • Chronic chlorate triggers functional senescence in vivo preceding overt neuronal loss. • Redox stress intensity shifts cellular response from senescence to acute apoptosis. • p21 serves as a redox-sensitive sentinel biomarker for waterborne neurotoxicity.
