• In stratum corneum, intercellular lipids form domains, and a domain should be constructed by one lamellar structure and one hydrocarbon-chain packing structure. • To solve the structural formation in a domain, differential scanning calorimetry and X-ray diffraction experiments as a function of temperature were performed. • Importantly, the short-period lamellar structure and the original orthorhombic hydrocarbon-chain packing structure are in the same domain, then the structures within this domain play a role in regulating the water content in stratum corneum.
