• Severe dengue infection induced hepatic injury and disrupted carbon metabolism in murine models. • Altered cell–cell communication between hepatocytes and HMPS disrupted carbon metabolism. • Fostamatinib significantly suppressed liver injury progression. • Fostamatinib alleviated inflammation and restored carbon metabolic homeostasis in SD mice.
