• Innovative synthesis of EM-CPS nanoclusters for inducing cuproptosis via low-dose RT. • Sulfur hybridization generates abundant ion pairs while enhancing triple enzymatic activity. • Exosome coating improves biocompatibility, tumor accumulation, and reduces off-target cytotoxicity. • In vivo studies demonstrate potent tumor suppression, robust immune activation effect, and excellent biosafty.
