• One-pot four-component domino cyclocondensation for pyrimidine-2,4-diones. • DES-functionalized silica acts as a recyclable heterogeneous catalytic system. • 2-MeTHF-mediated protocol under ambient conditions with high atom economy. • Rapid conversion (≤15 min) with excellent yields (up to 94%) and selectivity. • Antimicrobial screening reveals promising bioactivity of synthesized scaffolds.
