Brain development proceeds in a precise order, guided by transcription factor programs and constrained by epigenetic barriers that regulate chromatin accessibility and gene expression dynamics. Chromatin regulators establish specialized chromatin states that act as a developmental clock, enabling the stepwise activation of growth, differentiation, and maturation programs. Disruption of these chromatin-based mechanisms contributes to diverse neurodevelopmental disorders and drives pediatric brain tumors by generating cells that remain arrested in primitive, progenitor-like states. Identifying shared downstream pathways and epigenetic vulnerabilities can facilitate biomarker development and inform treatment strategies across diverse neurodevelopmental disorders and pediatric brain tumors.
