• EP300 and CREBBP encode two homologous acetyltransferases p300 and CBP. • EP300 and CREBBP are relatively frequently mutated in cancers. • Some of p300/CBP inhibitors emerge promising in anticancer approaches. • Simultaneous mutations in EP300 and CREBBP may impact efficacy of p300/CBP inhibitors.
