• Genome-wide identification of IL genes in Nile Tilapia. • IL family was expanded via tandem duplication and whole-genome duplication in tilapia. • Six ILs showed differential expression in spleen after S. agalactiae infection. • Specifically, IL-17A pathway activated the expression of IL-6, IL-8, and IL-8L in spleen cells. • And the promoter of IL-17D underwent demethylation in spleens after S. agalactiae treatment.
