• This is the first report to systematically characterize the PbrRLCK family and functionally link a specific member, PbrRLCK99, to disease resistance against the fungal pathogen Alternaria alternata. • PbrRLCK99, which is localized to the plasma membrane, acts as a positive regulator against A. alternata. Silencing of PbrRLCK99 suppresses resistance by reducing the activities of defense enzymes. • Meanwhile, overexpressing PbrRLCK99 boosts resistance by elevating the expression of salicylic acid (SA) marker genes and enhancing the activities of defense enzymes.
