There is a growing need for portable, lightweight, battery operated instruments capable of detecting and identifying bio-warfare and bio-terrorism agents in the field. To address this need, we have developed a handheld PCR instrument (Figure 1). LLNL s advanced thermal cycling technology and expertise with portable, field tested biological instrumentation, combined with the development of real-time, fluorescence based PCR assays, has enabled the development of a very portable, versatile, power efficient PCR instrument with a simplified operating system designed for use by first responders. The heart of the instrument is the sample module, which incorporates the advanced silicon thermal cycler developed at LLNL. The handheld unit contains four of these modules, each of which is capable of performing two simultaneous DNA assays. The new thermal cycler design has a novel anisotropically etched architecture that reduces thermal mass and increases cooling efficiency. This combined with a new thin film heater/temperature sensor enables extremely rapid and accurate thermal cycling. The system also incorporates an onboard temperature calibration system, which allows quick calibration while in the field. Each thermal cycler has two windows, allowing for two independent optical paths for excitation and detection. The sample module houses two LED excitation sources and interchangeable filter sets for both excitation and detection. This creates a flexible system allowing the use of real time Taqman (R) ratio assays or user defined two-plex assays.
