• Recombinant xylanase production was integrated for rice straw XOS. • Fed-batch E. coli cultivation yielded xylanase activity of 207.95 U/mL. • Mild alkaline pretreatment was optimized using response surface methodology. • Hydrolysis produced 6.13 mg/mL XOS, with X2 and X3 as dominant fractions. • Linking enzyme supply with pretreatment improved process consistency.
