• Cells co-expressing luciferase and red fluorescent protein were generated. • Luciferase measurements and cell count correlate, validating its application. • Luciferase activities were used to accurately quantify variations in cell invasion. • Red fluorescent protein dTomato provided real-time visualization of invasion. • Our established assay showed high sensitivity for measuring invading cells.
