• HDAC7 overexpression protects against hypertrophy in vitro and in vivo. • HDAC7 interacts with and represses MEF2D to suppress hypertrophic gene expression. • Our findings demonstrate the therapeutic potential of HDAC7 as anti-hypertrophic agent.
