Cells face constant challenges from a wide array of environmental and internal stresses that threaten their function and integrity. To maintain organelle function and cell viability in these situations, cells modify the activity of key components of membrane trafficking pathways and activate alternative transport pathways to bypass damaged or overloaded routes. This review highlights recent advances in the understanding how transport at the endoplasmic reticulum and the Golgi complex responses to various stresses to maintain homeostasis at the organellar and at the cellular level.
