• WGCNA identifies CTSS, NFKBIZ, and TNFAIP2 as hub genes in sepsis. • Hub genes are linked to neutrophil immunosuppression in septic patients. • scRNA-seq confirms hub gene expression in immunosuppressed neutrophil subsets. • The CLP model validates the role of these hub genes in septic immunosuppression. • Our findings suggest novel therapeutic targets for sepsis-induced immunoparalysis.
