• Sericin contributes to the formation of heterotypic 3D breast tumor spheroid formation. • Low share stress under orbital agitation supports stable and reproducible spheroid assembly. • The supplementation of 0.1% sericin has been demonstrated to enhance the compactness, sphericity, and integrity of spheroids. • Sericin-based 3D models enhance physiological relevance for drug testing and cancer research.
