Gene escape from X-chromosome inactivation can cause sex-biased diseases. A workflow is presented to help find escape genes and experimentally manipulate their expression. Sex-biased diseases may arise from differential expression of genes escaping X-chromosome inactivation in female individuals; however, causality remains mostly unproven. To help find escape genes that cause sex-biased disease, we present an in vivo workflow to pinpoint when and where escape matters, prioritize conserved candidate genes, and experimentally manipulate their expression.
