• MPC1 is a negative regulator of LPS-mediated inflammatory response. • Overexpression of MPC1 does not promote the formation of endotoxin tolerance. • Mitochondrial oxidative stress resistance promotes endotoxin tolerance. • MPC1 regulates mitochondrial oxidative stress resistance and endotoxin tolerance through the SIRT3/SOD2 axis.
