• A novel MHNF-DTI framework is proposed for DTI prediction. • The MHNF-DTI encodes drug structure and protein sequence simultaneously. • A multi-domain label reversal dataset is built to mitigate hidden ligand bias. • The performance of MHNF-DTI outperforms the existing state-of-the-art baselines.
