• A NiFe-LDH-CoTCPP heterostructure is constructed to suppress ACQ effect via in situ crystallization. • OER-induced HER process generates abundant ROS for multichannel ECL self-enhancement. • The synergy of OER and HER is pioneered for ECL amplification in a continuous potential scan. • An ultrasensitive ECL biosensor achieves miRNA-221 detection with a low limit of 16 aM.
