• Construction of a large-scale causal atlas linking the plasma proteome to osteoporosis. • Identification of two novel osteoporosis susceptibility loci (HOXC5 and HBQ1) via GWAS meta-analysis. • Revelation of three core pathological axes: WNT signaling, NUDT2-mediated metabolism, and pulmonary-bone crosstalk. • Characterization of NUDT2 as a druggable regulator of osteoprogenitor differentiation.
