• Systematically unravel O3-NFM elastic anisotropy evolution during desodiation via first-principles calculations. • JT-active ions occupancy modulates the phase-dependent elastic modulus degradation rate. • The evolving strength of intralayer (TM-O) and interlayer (Na-O) bonding during desodiation drives an exponential increase in anisotropy index.
