• Achieved the first heterologous synthesis of ergothioneine (EGT) in Aspergillus niger. • Identified Tr egt1 and Nc egt2 as the optimal genes enhancing EGT biosynthesis. • Precursor supplementation and fermentation optimization boosted EGT yield to 3.12 g/L. • Established A. niger as a promising fungal chassis for eukaryotic metabolic engineering.
