• Introduces a label-free Phase Correlation Spectroscopy (PCS) technique inspired by fluorescence correlation spectroscopy (FCS). • Utilizes time-resolved phase fluctuations from quantitative phase maps analogo08us to fluorescence intensity fluctuations in FCS. • Determines diffusion coefficients of micron-sized polystyrene microspheres. • Hydrodynamic radius estimated from PCS-derived diffusion coefficients matches the actual microsphere dimensions, confirming method accuracy. • Results follow Fick’s second law for concentration-dependent diffusion. • Demonstrates a non-invasive, label-free alternative to conventional FCS for diffusion analysis.
