• Stress-susceptible rats and stress-resilient rats were distinguished by sucrose preference test. Stress-susceptible rats exhibited downregulated neurotransmitter level and decreased presynaptic vesicle density compared to stress-resilient rats. • PCDH10, MANF, Septin 11, and UFD1 were downregulated in the prefrontal cortex but upregulated in the hippocampus of stress-susceptible rats, all of which are opposite in the stress-resilient rats. • Knockdown of PCDH10 in the hippocampus of rats produced a resilient phenotype, effectively ameliorating depression-like behaviors in rats exposed to chronic unpredictable stress. • Overexpression of PCDH10 in the hippocampal of rats did not directly induce depression-like behaviors but markedly increases susceptibility to stress, leading to exacerbated behavioral deficits after stress exposure.
