• Confined-free DNA nanosphere amplifier (CF-SCA) enables high-fidelity miR-21 sensing. • It solves the CHA dilemma between background leakage and sluggish reaction kinetics. • The enzyme-free platform achieves a 100 pM LOD with excellent nuclease resistance. • CF-SCA resists macromolecular crowding for dynamic in situ miRNA imaging in cells. • It successfully stratifies multi-cancer clinical serum without RNA extraction.
