• HPLC fingerprints identified 13 common peaks in raw and wine-processed Curculigo rhizome. • Wine processing enhanced bioactive compound content and reduced nephrotoxici potential compared to raw form. • Both extracts alleviated arthritis symptoms and synovial inflammation in CIA mice dose-dependently. • Spectrum-effect analysis revealed ANA, SAK, CA, and ORC as key anti-rheumatoid arthritis markers. • Network pharmacology and molecular docking identified glycolysis inhibition as potential mechanism.
