• αTAT1 is the sole α tubulin K40 acetyltransferase in zebrafish. • αTAT1 and α tubulin K40 acetylation are dispensable for embryonic development and cilia driven morphogenesis. • Cilia structure and motility are preserved in αTAT1 mutants despite complete loss of α tubK40Ac. • αTAT1 dependent acetylation is required for normal touch evoked sensory behavior in zebrafish.
