• High-precision imaging assays are applied to provide detailed insights into intracellular DNA trafficking. • Labeled DNA uptake is quantified with a novel anti-dye-antibody assay. • Colocalization of DNA with barrier-to-autointegration-factor (BAF) is assessed. • Imaging assays are used to study differences between protein-based TFAMoplex transfection systems and Lipofectamine. • TFAMoplex-based transfection mediates strong DNA cell association, high particle uptake and high transfection efficiency.
