• Chalcone derivatives 7a inhibited nitric oxide and pro-inflammatory cytokine production in LPS stimulated RAW 264.7 cells. • 7a suppressed osteoclastogenic activity in RANKL-induced osteoclast cells. • 7a downregulated the activation of NF-κB and MAPK signaling pathways involved in inflammation and osteoclastogenesis. • In-vivo, 7a reduced LPS induced systemic inflammation, carrageenan-induced paw edema and serum levels of CRP and LDH.
