• ZWINT expression is significantly upregulated in the COPD-NSCLC model and drives CD4+ T cell exhaustion and ferroptosis in vivo. • ZWINT directly induces mitochondrial dysfunction and ferroptosis in CD4+ T cells in vitro. • The tumorigenic effect of ALYREF depends on its regulation of the downstream target ZWINT.
