• TICT-based fluorescent probes were developed for selective detection of tau and Aβ aggregates. • Solvatochromism, Lippert–Mataga, viscosity, and DFT calculations confirmed the TICT photophysical behavior of the probes. • Probe 4a and 5d selectively detected tau and Aβ aggregates, respectively, in living cells. • Probe 5d selectively labeled Aβ plaques in mouse brain tissues with high specificity.
