• AMER1 loss in CRC correlates with advanced staging and CD8+ T cell exclusion. • AMER1 physically binds and shields DDC from degradation to drive dopamine synthesis. • Tumoral dopamine accumulation triggers GSDMD-dependent cancer cell pyroptosis. • Dopamine therapy reverses CD8+ T cell terminal exhaustion to halt CRC progression.
