• A single-cell culture microchamber array chip was developed for drug-resistance evaluation. • It enables stable single-cell trapping and culture in a microchamber array designed to allow neighboring cells to remain in close proximity. • The ability of individual cancer cells to proliferate in anticancer drug–containing medium was evaluated.
