• Aromatic ring-cleaving dioxygenases catalyze O-heterocycle cleavage. • CatA and MhpB cleave the O-heterocycle 2-hydroxytetrahydrofuran (2-OH THF) in vivo. • E101 (CatA) and H115/R212 (MhpB) contribute to 2-OH THF cleavage activity. • catA redundancy and active site conservation indicate adaptation to mixed pollutants.
