• The VGCC auxiliary subunit α2δ-1 is a putative GluA1 ATD interacting protein. • Postsynaptic α2δ-1 deletion increases basal synaptic AMPAR transmission. • Presynaptic α2δ-1 deletion reduces LTP without altering basal synaptic transmission.
