• A surface-enhanced Raman scattering substrate consisting of reduced graphene oxide encapsulated in molybdenum disulfide with additional Ag nanoparticles (Ag NPs) decorated on the surface was developped. • The developed RGO@MoS2/Ag NPs SERS substrate enables the highly sensitive, label-free detection of IL-6. • The developed SERS substrate achieves a detection limit as low as 10-9 M, with an enhancement factor of 2.2 × 109. • The SERS substrate detected IL-6 with a good linear response in the 5.0–1000 pg/mL range and a detection limit as low as 0.41 pg/mL.
