• A dual-mode ECL/SERS biosensor was developed for sensitive APE1 detection. • An X-shaped DNA recognizer enabled efficient APE1-triggered cascade amplification. • A PAM interface integrated ECL emission and plasmon-enhanced Raman readout. • APE1 induced opposite ECL decrease and SERS increase for self-validation.
